The PTC (PhenylThioCarbamide) is a bitter organic compound found in certain plants such as Brussels sprouts or broccoli, about 30% of the population is sensitive to this compound. The objective of this practical work is to propose to the students to establish the relationship between phenotype and genotype. The phenotype is easily characterized using test strips. The genotype is determined by gene amplification of a portion of the TAS2R38 gene from two provided DNA fragments (plasmids): - One plasmid contains the version of the TAS2R38 gene that confers sensitivity to PTC. - One plasmid contains the version of the TAS2R38 gene that confers non-sensitivity to PTC. PCR is followed by enzymatic digestion and electrophoresis gel migration. This new protocol allows conducting the experiment on a synthetic gene without sampling from humans! Several scenarios are available to you!
