In the world, more than 460 million people have diabetes, a disease that increases the risk of death. Type 2 diabetes (T2D) represents 90% of diabetes cases. The creation of murine animal models is necessary to study this disease. Mutations in the LEP or LEPR genes lead to early obesity and hyperglycemia accompanied by insulin resistance in mice, with reported mutations in humans as well.
Using the CRISPR-Cas9 system, it is possible to generate mouse models with mutations that make correct protein synthesis impossible. These mice are obese and display a diabetic phenotype from the first generation. The genotypes of these LEP and LEPR KO (gene-deficient) mice are confirmed by PCR and DNA electrophoresis.

This kit allows for simulating a search for mutations induced by CRISPR-Cas9 in these mice and establishing the link between their genotype and the diabetic phenotype.

The DNA solutions are calibrated and require no preparation.
