Cystic Fibrosis Kit (DNA electrophoresis)
Cystic fibrosis is a severe genetic condition characterized by an increase in the viscosity of bodily secretions, particularly affecting the respiratory and digestive systems. Over 1500 identified mutations in the CFTR gene alter the function of the CFTR transmembrane protein synthesis. The activity is presented in the form of a pre-implantation diagnosis (fictional). Parents wishing to have a child through IVF undergo this diagnosis due to family history. This lab practical follows the principle and format of the CF4 medical test (Zeneca diagnostic), a test based on PCR ARMS amplification. It identifies the 4 most common mutations of the CFTR gene: DeltaF508 / G542X / G551D /621+ through electrophoresis. The principle involves a dual analysis where a healthy gene is sought in one tube and its mutated allele in the other tube. Each individual has 2 tubes containing the PCR products, determining their genotype based on the presence or absence of one or more mutations. The DNA solutions are calibrated and require no preparation.
