Kit Influence of auxin and cytokinin on the development of the carnation

Principles and objectives:
The experiment consists of studying the influence of the nature of the hormones present on the type of growth obtained.
In vitro culture is carried out from a node of a carnation plant. The seeds must be germinated at least 6 weeks before the lab session to obtain plants of sufficient size.
The growth results observed on the three media are the formation of a plant, a callus, and the rooting of the seedling, after 4 to 6 weeks of culture.
To carry out this experiment, plan for 3 sessions of 50 minutes each. Reagents are provided for 10 to 20 tests depending on the flask used.
Composition:
- 1 packet of carnation seeds
- 1 flask of Medium OE1 enriched with Cytokinin (dose for 400 mL)
- 1 flask of Medium OE2 enriched with Cytokinin + Auxin (dose for 400 mL)
- 1 flask of Medium OE3 enriched with Auxin (dose for 400 mL)
Additional materials needed: flask with cap or glass test tube, absorbent cotton, sterile forceps, scalpel, 95° ethanol, water bath, Bunsen burner or electric burner, mini greenhouse
Comments:
We have described in the instructions the entire protocol to follow to successfully carry out the experiment.
At the end of the lab session, the rooted seedlings can be transferred to a greenhouse.
