Use the pGLO bacterial transgenesis kit to teach gene expression and genetic engineering. Explore the concepts DNA > RNA > Protein > Phenotype and the roles that internal and external factors play in gene regulation.
 
In this activity, students use the pGLO plasmid to transform bacteria to express the Green Fluorescent Protein (GFP) from the jellyfish Aequorea Victoria, which makes the bacteria glow green under UV light. Bacteria transformed with the pGLO plasmid exhibit green fluorescence only when grown in the presence of arabinose. Use this experiment to illustrate that not all genes are expressed at the same time and that environmental factors control when gene expression occurs.

Students will carry out a complete transformation protocol identical to the protocol used daily by scientists around the world.


Experiment Duration:  Two 45-minute sessions
