Euglenas delivered in a transparent glass bottle of 50 mL.
Upon receipt, euglenas can be kept for 2 weeks, in the open air (cap unscrewed). For proper storage, and mandatory if the desired storage duration is longer than 2 weeks, it is advisable to culture the euglenas in a medium without lactic acid (ref. 107527). This is a concentrated medium, to be diluted 10 times.
To maintain the strains:
*Work under the most sterile conditions possible
- In a sterile 500 mL flask, pour the lactic acid-free medium 10X bottle (107527) and complete with sterile distilled water up to 500 mL.
- In another sterile flask (250/500 mL), pour all of the microorganism suspension bottle and close with a cotton ball wrapped in gauze (puppet).
- Add between 20 and 50 mL of medium every 2 weeks and shake the culture flask very regularly (ideally daily). Expose to light for 12/14 hours a day at a temperature above 18-20°C.
Photosynthesis Lab:
Euglenas are mixotrophic organisms. They can switch from autotrophic to heterotrophic behavior.
- To promote their growth, they are cultured in a medium with lactic acid (complete). The external food supply allows euglenas to multiply rapidly.
- If you wish to conduct an experiment on photosynthesis in ExAO, it is advisable before the lab to transfer the euglenas to a medium without lactic acid (carbon source - ref. 107527) in the presence of light to wean them off and starve them, thus encouraging them to move away from heterotrophy. Start measurements in the bioreactor with a dark phase.
